ABSTRACT:
The study was aimed to develop a genistien (GN) loaded nanoemulsion based nano-gel in order to ensure increased photo-protection efficacy against UVA induced oxidative stress and its enhanced bioavailability through transdermal delivery. The optimized GN-NE2 composed of an appropriate ratio (15:30.55 % w/w) of the oil (labrafac Lipophile WL1349), water and Smix (surfactant (labrasol) and co-surfactant (PEG 400)] respectively. GN-NE2 was characterized by through droplet size, zeta potential, refractive index, TEM, UV and FTIR analysis. Ex vivo skin permeation and in vivo UVA protection activity of GN-NE2 based nano-gels (GN-NG2) along with placebo-NG were studied through the rat skin. The pharkacokinetics and bioavailability of pure GN and its nano-gel, GN-NG2 was studied at dose of 20 mg/kg in rats. GN-NG2 exhibited enhanced UVA protection efficacy and antioxidant potential due to its improved pharmacokinetics (Cmat: 95.06 z 4.09 ngml"; tuzel: 3.81 2 0.25 h) and % relative bioavailability (352.58) when compared to pure GN. Nano- gel significantly ('P < 0.01) released the drug in a sustained manner to maintain the minimum effective concentration for extended period of times. Nano-gel could elevate the level of skin marker enzymes against oxidative stress mediated by UVA. Thus, transdermal route of nano-gel of GN offers an effective strategy for enhancing their bioavailability.
