Abstract: Hairy root
cultures are induced from Agrobacterium rhizogenes mediated transformation
technique. Transgenic hairy root is an optimistic source of secondary
metabolites in medicinal plant with high pharmaceutical value and for the
production of phytochemicals or medicinally important enzymes and foreign
proteins either in in-vitro conditions or through plant regeneration. The hairy
roots are grown in vitro in bioreactors to study their soil interaction with
other pathogens like fungi, nematodes and to study the mass production of
various secondary metabolites. So, this technique is a powerful tool in the
field of ethno pharmacology. The study emphasized on the development of a
reliable and well defined protocol for the induction of hairy root through
Agrobacterium rhizogene mediated transformation in Vigna radiata. Vigna radiata
can be used as antioxidant in treating various conditions like liver disease,
cancer, diabetes, kidney disorders and for curing various microbial infections.
This process is divided into three phases: first maintenance of Agrobacterium
rhizogene (MTCC 532) culture in different culture medium like Nutrient Broth
(NB), Yeast Extract Peptone (YEP) and Yeast Mannitol Broth (YMB), second was
in-vitro germination of different seeds has been observed, Vigna radiata was
showed highest rate of germination and third was initiation of hairy root
culture in different Vigna radiata 2nd day seedlings was showed 70% transformed
roots.
