In Vitro Clonal Propagation of Superior Line of (Trichosanthes dioica, Roxb.) an Economically Important Plant

Abstract: Pointed gourd (Trichosanthes dioica Roxb.) belongs to family Cucurbitaceae, common name Parwal the plant is a perennial, dioecious, and grows as a vine. Traditionally T. dioica is multiplied through seeds, stem cuttings and root cuttings. Propagation from seeds is not advisable due to less germination percentage and imbalanced male-female ratio. Protein and minerals were rich in fruits and are used in traditional system of medicine since ancient times. Pharmacological activates of T. dioica is haemagglutinating, antibacterial, antifungal, hypoglycaemic. An efficient protocol was developed for micro- propagation of superior line of an economically important, a perennial vegetable crop. Shoot initiation of T. dioica derived from green house grown plants, nodal segment (1.5-2 cm) were inoculated in culture tubes containing semisolid MS medium supplemented with 0.5 mg/ BAP. After 4 week, the cultures were inoculated in second medium for shoot multiplication containing MS medium supplemented with 1.0 mg/l BAP. The cultures showed significant increase in shoot length and shoot number. Maximum (90%) shoot elongation with an average shoot length of 4.4+0.06 cm was observed at 1.0 mg/ BAP. To further enhance the number of shoots per explants, mother tissue was repeatedly subculture on fresh shoot induction medium after each harvest of newly formed shoots. Thus, by adopting this strategy, an average of 53 shoots per explants could be obtained. After the root formation, the in vitro raised plantlets removed from culture tubes, washed thoroughly in running tap water and then were planted in small root trainers containing vermiculate for hardening in green house under temperature 29+2°C and relative humidity 70%. After fifteen days of transfer, 75 percent plants survived in the green house.
